Introduction
Langerdorff heart or also known as isolated perfused heart is an in vitro research that used animals such as rat, guinea pig, rabbit and cat as the subject to examine or study the heart rate and contractile of strength of the heart. In this experiment, the animal’s heart is being removed from its body and transferred to a constant pressure perfusion apparatus. The heart is allow to adjust to the new surrounding or environment for 15 to 30 minutes. Then, several perfusion fluid that is oxygenated, equilibrated to pH 7.4 and filtered is being test to the heart. Advantage of using this method is it lowers and reduces possibility of myocardial damage as a result of ischaemia and is a mehod of choice for the removal of heart from a freshly killed animal. First, a small balloon is inserted and fixed to a catheter tip and filled with water, into the left ventricle through one of the pulmonary veins, mitral valve and left atrium. Then, the end of the catheter is attached to a pressure transducer. Expanding the balloon will result in optimizing of ventricular preload. Second technique is by attaching or inserting a small hook a spring-loaded clip to the ventricle and connect it to a balanced isotonic lever (tension transducer). In order to take heart rate measurement, a rate meter or tachograph which are triggered from tension transducer is needed which will eventually will give a graph that illustrate the effect of fluid to heart rate. Next component that need to be measured is Coronary Blood Flow (CBF). CBF is the mean volume of perfusion fluid draining from the heart when the heart is perfused at constant
